Yes, DEP will work fine even if you haven’t turned on pre-enrollment.

Actually, if you turn on pre-enrollment, then the devices that are enrolled in DEP will be displayed on your portal only after the user completes enrolling the device. Turning pre-enrollment off will add every device to the list of DEP devices, even if it isn’t enrolled.
